That is a question that needs a little more information. Assuming that you are talking about (1) 220Ah 12Volt "True Deep Cycle" battery and (2) 220Ah 6Volt "True Deep Cycle" batteries in series, there should no difference. The 12VDC battery cells will be the same thickness as the 6VDC battery cells. BUT the size of the 12Volt battery will be close to putting 2 6Volt batteries side by side. 220Ah is 220Ah (of course you can only use 110Ah of the 220Ah - the 50% rule)
So when you want to compare you need to compare a simuilar rated 12Volt battery to a pair of simuilar rated 6Volt batteries.
BUT, most 12VDC batteries are a lower Ah rating, so 2 6's would be better if an equal 12Volt one of equal Ah could not be located.
