Seams a standard the "main black tank" has the flush connection and the "Aux black tank" will not. We carried a flush wand to clean the aux black tank. You'll need an additional hose but well worth it.

I have a '19 29BHDB, each black tank has has its own flush. But, you're asking how to know they're connected. I would hook each up (one at a time) with a dedicated "black tank hose". Open the black tank valve with your dump hose connected (at the dump preferably) and then turn on the water for the flush. You should start hearing water move through the sewer hose. If not, it's going somewhere else and I think would become evident from leaking under the trailer after a couple of minutes.
