Water heater

In order for the water heater to fill you must make sure it isn't in winter bypass mode and then open a hot water tap somewhere (inside sink, outside shower, pressure relief on the water heater) to let any air escape. When the water flows freely your tank is full
Do not, I repeat, do not turn on the water heater burner or electric element until the tank is full.
Another tip is to make sure the water heater is full before you leave for a boondocking trip. Thats 6 or more gallons that would be sucked out of the fresh water tank.
 
When you hookup to city water does it automatically fill up the water heater

I'd add: The valves used to bypass the hot water heater for winterization purposes are typically located next to the hot water heater itself (on the inside of the trailer). These 2 or 3 valves need to be adjusted so that incoming water flows through the hot water heater vs. bypassing it. The water from both the trailer's on-board freshwater tanks and the city water connection flow through these valves.

To clarify, water will still flow through the hot water side of the faucets even if the the hot water heater is empty and still in bypass mode. The best check to make sure the hot water tank is full of water and not bypassed (as Dobbs also mentioned) is to make sure water (and no air) flows freely (under pressure) from the hot water heater pressure relief valve. This valve is located behind the hot water heater maintenance door on the outside of the trailer.
