To expand on what ALJO said: The primary goal is to prevent cool air from escaping from the cool air plenum into the space between the ceiling and the plenum.
When you remove the duct from the ceiling, there will be a short section of rigid plastic tubing. By removing (discarding) that tubing and taping the space, it will prevent air from escaping AND allow easier air flow from the plenum into the ceiling duct.
We did this mod earlier this year and there was a significant increase in air flow from one of our 3 ceiling vents at the rear of the trailer. I believe it was due to the plastic tube being too close to the top of the plenum and restricting air flow into that ceiling vent.
